This is an exciting opportunity to develop cutting-edge and diverse high-throughput functional genomic and CRISPR screening technologies to aid in the discovery and advancement of early-stage therapeutic targets in the context of solid tumor cancer resistance mechanisms. The selected candidate will join the Early Target Biology group within the Mechanisms of Cancer Resistance Thematic Research Center at the new Cambridge Crossing site. The site is highly collaborative and the candidate will have exposure to all stages of drug discovery as part of their growth and development.
Core Job Functions - Develop and implement high-throughput, functional genomics screening assays (such as genome-wide CRISPR screens, digenic screens, protein tiling screens) with potential to implement advanced genomics technologies as part of a highly collaborative project team.
- Validate novel targets arising from screening efforts using a variety of mammalian cell-based assays
- Collaborate with data analysts and project teams across asset progression as well as in the translational research space
- Interface and coordinate with academic collaborators. Keep up to date with the latest state-of-the-art technologies in the genomics/CRISPR space.
- Organize, track and progress projects in a timely fashion.
- Independently design experiments, compile and analyze data, troubleshoot novel methods as needed, and maintain lab notebooks, protocols, and written records.
- Train and assist other team and site members.
Basic Qualifications - Bachelor’s Degree
- 5+ years of academic and / or industry experience
Or
- Master’s Degree
- 3+ years of academic and / or industry experience
Or
- Ph.D. or equivalent advanced degree in the Life Sciences
Preferred Qualifications - Strong scientific and hands-on experience with:
- High throughput gene perturbation screening techniques (such as whole genome or targeted CRISPR screens, Perturb-seq, CRISPR tiling screens, shRNA screens, deep mutational scanning, ORF screens, digenic screens, flow cytometry-based screens)
- Genome engineering techniques (gene knockout/knockdown/knock-in)
- Mammalian tissue culture, lentivirus generation, viral transduction, and transfection
- Cellular and molecular biology techniques such as RNA/DNA isolation, western blotting, flow cytometry, RT-PCR, cell proliferation assays (CTG, incucyte), molecular cloning and vector design
- Proficiency in utilizing software such as ELN, Graph Pad, Microsoft office etc. for data analysis and figure generation is required.
- Solid tumor cancer biology, CRISPR guide design, vector optimization, NGS library construction, familiarity with NGS data analysis and interpretation, high throughput assays and automation, cancer genome data mining (eg. cBioportal, Depmap)
- Attention to detail with excellent organizational and record keeping skills
- Excellent interpersonal skills with the ability to interact effectively with people, internally and externally
