The position
A key feature of next-generation rAAV-based gene therapies is the targeted expression of transgene in a cell type- or disease state-specific manner. This allows to detarget undesired tissues and cell populations to increase safety and thus expands the therapeutic space of rAAV gene therapies in general.
As a Roche RPF Postdoctoral Fellow (m/f/d) in our Nucleic Acid Based Medicine (NABM) matrix team, you will focus on an exploratory research project related to the development of novel gene regulatory elements, which allow precise regulation of therapeutic transgene expression. To achieve this goal, you will identify, functionally validate and optimize novel genetic elements in alignment to current and further portfolio needs at Roche pRED. This gives you the opportunity to collaborate and connect to different functions internally as well as to external academic partners.

Main task of this position:
You will utilize state of the art computational tools (in R or Python) to analyze large datasets in order to identify endogenous regulatory DNA sequences
You will establish and perform NGS based high-throughput screening assays to functionally characterize DNA sequences
You will employ machine learning approaches to build fully synthetic regulatory elements in close collaboration with our computational department
You will manage and organize the experimental workstream between different wet and dry lab teams.
This position is sponsored by the Roche Postdoctoral Fellowship Programme. The RPF Programme has the ambition to build and maintain the Scientific Leadership of the future and to provide leading Roche scientists with the opportunity to collaborate with talented postdoctoral researchers, and leading academic research groups, from around the world. The duration of the RPF project is initially set for two years, with the possibility of extension for a third year. You will be based in Penzberg within the greater Munich area (Germany).

Essential skills and expertise:
Experience in design and cloning of large DNA barcoded libraries
Understanding of transcriptional and translational gene regulation
PhD in molecular biology
Self-organized, autodidactic and creative mindset to establish novel high-throughput screening approaches

Preferred skills and qualifications:
Generation of costume NGS sequencing libraries
Basic computational experience in either R or Python
Experience in generating and working with different multiomic or epigenetic datasets
Fluent communication skills with both computational and biological scientists in English

We anticipate the start date of this fellowship to be mid-2024, by which point you should have successfully handed in your PhD thesis or no later than 5 years after the PhD.
